While every site contains unique content and graphics, you may have noticed that
there are 3 primary layouts used in most of today's Websites.
Most sites are have either a 1, 2, or 3 column layout design.
Deciding which one is right for you depends mostly on what type of site you have and the amount of information contained in your site.
The more sections you have, and the more information your site contains, the more important your choice of layout type becomes. This is part of the information architecture of the site.
The following are screenshots and a brief description of some of the functionality provided on a few of the sites I have designed (I also host and maintain these sites).

The Texas Society of Anesthesiologists Website is an example of a non-profit organization site: http://www.tsa.org/

The Health Resource Website is an example of a small business site: http://www.thehealthresource.com

Kinetic Arts Stained Glass Studio Website is another example of a small business site: http://www.kineticarts.org/

Ryan's Tree for Grieving Children Website is an example of a small non-profit organization brochure style site.
Ryan's Tree non-profit became associated with a local hospital and the site is no longer active.
